ADP-Glo™ Kinase Assay is a luminescent kinase assay that measures ADP formed from a kinase reaction; ADP is converted into ATP, which is converted into light by Ultra-Glo™ Luciferase. The luminescent signal positively correlates with kinase activity. The assay is well suited for measuring the effects chemical compounds have on the activity of a broad range of purified kinases, making it ideal for both primary screening as well as kinase selectivity profiling. The ADP-Glo™ Kinase Assay can be used to monitor the activity of virtually any ADP-generating enzyme (e.g., kinase or ATPase) using up to 1mM ATP.

Figures

Principle of the ADP-Glo Kinase Assay.Figure 1. Principle of the ADP-Glo™ Kinase Assay.

The assay is performed in two steps: 1) after the kinase or ATPase reaction, ADP-Glo™ Reagent is added to terminate the kinase reaction and deplete the remaining ATP, and 2) the Kinase Detection Reagent is added to convert ADP to ATP and allow the newly synthesized ATP to be measured using a luciferase/luciferin reaction. The light generated correlates to ADP present and kinase or ATPase activity.
